Latency in Morris water maze analysis

Latency, the time taken for the subject to reach the hidden platform, is one of the most commonly used measures in Morris water maze analysis. It’s a very simple index of learning and performance – which both makes it valuable and gives it limitations.

Latency is a direct, scalar measure that is simple to record, giving a single number summary that is easy to compare across groups. It gives clear, time-based readouts that reflect changes in learning and performance, including impairments.

  • In cue-learning (visible platform) trials, short latencies show that the subject can see the cue and swim toward it and that the animal understands that finding the platform ends the trial. If latency does not improve over repeated cue trials, it could indicate visual or sensorimotor impairments, lack of motivation or general cognitive issues.
  • In repeated hidden-platform trials, a consistent drop in latency indicates successful learning of the platform location, which often reflects acquisition of spatial memory (but see below).
  • In reversal learning trials, changes in latency (decreasing before platform relocation then increasing when the platform is moved, then decreasing again or not) can reflect cognitive flexibility in adapting to the new location or difficulty in suppressing old spatial memories.
  • In probe trials, latency to the former platform location (if measured) can provide supporting evidence of spatial memory, though is rarely used as a primary measure.

There are, however, important limitations to consider:

  • Latency does not reveal strategy. It doesn’t tell you whether the animal used a spatial strategy, a procedural one (e.g. chaining), or found the platform by chance. A low latency might for example reflect habitual motor sequences developed through overtraining, rather than use of a spatial map. This is particularly important in distinguishing between hippocampal and striatal strategies.
  • It’s affected by motor speed and activity levels. Slow-moving animals (due to age, sedation, injury, or low motivation) may have longer latencies despite knowing the platform location; hyperactive animals may reach it relatively quickly even with poor navigation.
  • It’s biased by start point and start-goal distance: latency varies with different start positions, bringing non-learning-related variability into your data.
  • Latency may be insensitive to differences between animals or groups when scores are clustered at the extreme ends of the scale. A ‘floor effect’ may occur when highly trained animals find the platform very quickly, often within a few seconds, so latency can’t decrease any further and can’t detect fine-grained differences in performance or learning across subjects or treatments. Conversely, a ‘ceiling effect’ may occur when subjects don’t find the platform at all, or take nearly the maximum allowed trial time (e.g., 60 seconds), which is common in early training, severe cognitive impairment, or low motivation, where latency may not distinguish between partial spatial learning (e.g., focused searching near the goal) and non-specific or random behavior.

It’s therefore important to use additional measures such as path efficiency ratio, heading angle, Gallagher proximity measures, other measures and analyses and/or HVS Image’s automatic behavior classification, and not to rely on latency as the primary measure.

While latency is sometimes still measured manually, automated systems like HVS Image provide greater objectivity, precision, and reproducibility, giving a consistent, objective measure and synchronising latency with path length and other measures.

Depending on your experiment and personal preferences, you may want the time to end automatically when the system objectively determines that the subject has reached the platform, or you may want to judge this for yourself.

  • The time begins when the experimenter clicks the handheld remote as they release the subject into the pool, avoiding false starts that can be recorded by other systems (e.g. when the experimenter’s hand enters the pool area, before the subject is released).
  • For the end time you set one of the following options (with one click in the software):
    • Auto-stop on platform detection. This gives objective measurements and includes instances where the subject briefly climbs partly onto the platform.
    • Auto-stop after a fixed duration on the platform (you set the time, e.g 1 second). This gives an objective measurement and ignores instances where, for example, the subject briefly or partly climbs onto the platform and then continues swimming.
    • Manual stop by clicking remote. This allows the experimenter to end the time when appropriate, which may be particularly useful in unusual or unpredictable trial situations or non-standard protocols.
